Managing a gradual increase in back pressure over a column's lifetime
A moderate increase in back pressure is expected over the lifetime of the column. When this gradual increase of back pressure is experienced, it generally does not affect performance. This can be reduced, although not eliminated, by using digest techniques such as solid phase digestion (i.e. SP3) or FASP combined with stagetip desalting to minimise the amount of lipids and metabolites that are injected onto the column.
